全军创伤修复与组织再生重点实验室

-创伤感染与免疫学实验室简介

创伤感染与免疫学实验室(平台)是全军创伤修复与组织再生重点实验室和北京市皮肤损伤修复与再生医学重点实验室的重要组成部分。主要从事创(烧、战)伤后感染并发症,包括严重脓毒症、脓毒性休克与组织损害发病机制和诊治策略的转化研究。主要领域涉及细菌内/外毒素致病机制、免疫功能障碍及其监测、神经内分泌调节途径、脓毒症诊治新技术研究等。

该实验室主要学术成绩包括:(1)系统阐明高迁移率族蛋白B1在创、烧伤脓毒症发病中的确切作用与信号机制,首先提出其作为关键免疫调控分子诱发多器官损害的新观点。(2)发现生物喋呤参与内/外毒素休克的病理生理过程,建立了新喋呤和内毒素的临床预警阈值。(3)揭示了烧伤后金黄色葡萄球菌及其外毒素致病的临床意义,在国际上最早报道杀菌/通透性增加蛋白对肠源性内毒素血症具有重要防护效应。(4)提出评价严重烧伤免疫功能障碍程度的新标准,采用新的炎症、免疫调理策略对严重脓毒症进行干预,多中心试验疗效显著优于“国际脓毒症指南”推荐的方法。

实验室紧紧围绕以解决降低平时和战时严重创(烧、战)伤病死率中的关键科学问题开展研究,经过老中青三代近30年的建设、发展与凝练,逐步形成了严重脓毒症和脓毒性休克发生机制与诊治新途径的研究特色与学术优势。以盛志勇院士为代表的老一代科学家开创事业并奠定了坚实基础,以国家杰出青年科学基金获得者姚咏明研究员为代表的新一代专家推动和提升了科研团队的创新发展。该研究方向现有固定工作人员为7人,其中正高职1名、副高职3名,每年在读博士后、博士/硕士研究生以及客座研究人员20余人。姚咏明研究员为该研究方向负责人,清华大学、中山大学、南开大学等8所高校客座或兼职教授。目前担任国际休克学会执委、欧洲休克学会顾问、Shock杂志编委等。获欧洲、美国休克学会颁发的Schlag纪念奖、中国青年科技奖、“求是”杰出青年奖、“新世纪百千万人才工程”国家级人选、“全国优秀科技工作者”和军队杰出专业技术人才奖,入选“科技北京”百名领军人才工程等。主要学术骨干包括:(1)张庆红,美国NIH博士后,副研究员,主持国家自然科学基金面上课题2项和军队“十二五”计划重点课题1项。(2)祝筱梅,博士后,副研究员,北京市科技新星,主持国家自然科学基金课题2项;(3)董宁,医学硕士,副主任技师,研究团队的主要技术骨干。

实验室成员承担国家973项目课题、国家杰出青年科学基金、国家自然科学基金重点项目、国家自然科学基金面上和青年项目、军队杰出中青年人才基金、军队“十二五”计划重点课题等37项;在国内外专业期刊发表学术论文480余篇,其中被SCI收录112篇;主编大型学术著作5部,其中《现代脓毒症理论与实践》(177万字)、《急危重症病理生理学》(235万字)分别为国内相关领域首部大型著作;共获国际学术奖3项、国家科技进步二等奖4项、军队和省部级科技进步一等奖4项、二等奖8项。

该研究方向(平台)主要功能单元包括:分子与细胞免疫学研究平台、细菌内/外毒素检测技术、神经生物及神经行为学分析、基因组学与蛋白质学研究平台等。

Brief Introduction about Laboratory of Infection and Immunity

 

The research fields in this laboratory are focused on infection and immunity after severe trauma, burns, and war wounds, including the effect of hemorrhage and trauma on endotoxin response and its regulatory mechanisms, inflammatory cascade and subsequent sepsis induced by gram-positive bacteria and its exotoxin, and application of novel markers for diagnosis or prognosis in evaluation of organ functions as well as clinical course in septic complications. With both animal experiments and multicenter clinical trials, the ongoing study will be performed to investigate the potential mechanism and clinical significance of immune dysfunction in the pathogenesis of sepsis after severe burns and trauma. New theories concerning the criteria of different degree of immune dysfunction, and novel strategies would be formulated for the treatment of acute inflammation and immune regulation, in order to apply advantageous interventional measures.

We are greatly indebted to academician Zhi-Yong Sheng, the founder and academic guidance of the Laboratory of Infection and Immunity. The chief investigator of this laboratory is Prof. Yong-ming Yao, who is the winner of National Natural Science Outstanding Youth Foundation of China. A scientific research team is organized in the laboratory, with old experts in guidance, excellent young and middle-age staff as mainstay, and research staff of productive age and education as the structure of research staff.

At present, the laboratory is staffed by 7 scientific researchers and technicians, including 1 professor and 3 associated professors. The laboratory also enrolled more than 20 postdoctoral, doctoral, master graduate students and visiting researchers every year. In recent years, catching up with the international research progresses, the laboratory from time to time adjusted its research direction and obtained prominent progress and honor, such as the Schlag Memorial Award of International Shock Society, Qiushi Outstanding Youth Award, China Youth Science and Technology Award, New Century State Candidates for "Thousands Talent Project", Distinguished Young Scholars Supported by the National Science Fund, etc.

Since the establishment of the laboratory, we have published more than 480 scientific articles, with 112 by SCI source periodicals, and won 4 second class State Science and Technology Advancement Award. In addition, the members of this laboratory published 5 scholarly monographs as chief editor and 26 as editor.
